Thermoplasma Cultivation

Thermoplasma cells are polymorphic, varying in shape from spherical (0.3–2μm) to filamentous structures. It reproduces by budding. Its cells lack a true cell wall, but are surrounded by a single three-layer membrane, about 10-12nm thick. The genus is not motile. Colonies on solid medium containing 1.2% ionic agar, up to about 0.3 mm in diameter; these colonies are somewhat flat, coarsely granular, and dark brown. Some of them exhibit a typical "fried egg" appearance with a zona pellucida around the perimeter. Grow in liquid base containing 0.02% (NH4)2SO4, 0.05% MgSO4, 0.025% CaCl2·H2O, 0.3% KH2PO4, 0.1% yeast extract and 1.0% glucose. At concentrations below 0.2%, cell harvest was proportional to the concentration of yeast extract; no growth was observed on the ether extract fraction of yeast. During the cultivation process, sterols are not required. Thermoplasma are strictly aerobic bacteria. The optimum temperature for its growth is about 59°C. Grow well between pH 0.96 and 3.0; pH optimum is about 1-2. Thermoplasma is a true saprophytic bacteria that has been isolated from a pile of burned cinders and acidic hot springs.
