Cosmetic-grade Polyglutamic Acid

Cosmetic Grade Polyglutamic Acid (PGA) is a naturally derived amino acid polymer. Similar to hyaluronic acid and soluble collagen, PGA is an excellent hydrophilic humectant capable of forming a smooth, elastic film on the skin, thereby improving sensory perception, protecting the outer layers of the skin, and increasing the production of natural moisturizing factors.

Cosmetic Grade Polyglutamic Acid Products Options

Specifically designed for skincare formulations, it excels in enhancing moisture retention, surpassing conventional hyaluronic acid in certain aspects. As an effective cosmetic ingredient, PGA significantly improves moisture binding, elevating its efficacy in various skincare products. This can be very beneficial for stubborn dry, coarse, and flaky skin. PGA offering various molecular weights, each serving distinct roles in skincare. With a molecular weight around 5k Da, PGA ensures easy skin penetration and absorption, while PGA with a low molecular weight effectively combats free radicals, repairs photoaging, and rejuvenates the skin. On the other hand, higher molecular weights (ranging from 700k to 1000k Da) enhance water retention, ensuring optimal skin moisturization. These varied molecular weights cater to specific skincare needs, providing multifaceted benefits. Cosmetic-grade PGA is most commonly in the 500 kDa – 1,000 kDa range.

Molecular Weight Range Characteristics Cosmetic Application
PGA-SLM <10KDa Lower molecular weight sodium polyglutamate, better skin penetration, natural moisturizing factors accumulation, hydration capacity enhancement, melanin synthesis inhibition, and skin whitening. Lightweight serums, fast-absorbing gels
PGA-LM 10-700 Kda Low molecular weight, skin smoothing, natural moisturizing factors accumulation, water holding capacity enhancement. Synergize with HA, and improve skin elasticity and softness. Serums, masks, essence
PGA-HM >700KDa Slightly lower than super-high molecular weight, better film-forming moisturizing and thicken properties. Moisturizers, sheet masks, barrier creams
PGA-SHM ~2000KDa Molecular weight up to 2 million Daltons, long-lasting moisturizing film on the skin surface, resisting damage from external pollutants. Occlusive creams, sleeping masks

Optimized Production Platform

Produced through our high-density fermentation process using microbial hosts, the final product is pure and completely animal free. We have a range of products available including raw materials, finished forms and OEM services.

Polyglutamic Acid Production

Microbial production of cosmetic-grade PGA involves common or engineered microorganisms, typically Bacillus subtilis and Bacillus licheniformis, to express higher yields or enantiopurity (e.g. L-PGA vs DL-PGA), which are Non-pathogenic and GRAS (Generally Recognized as Safe). CRISPR/Cas or traditional mutagenesis strategies are utilized to boost PGA synthase (pgsBCA operon) expression and reduce degradation (deletion of pgdS).

Fermentation Medium Fermentation Conditions Downstream Processing
Parameter Typical Range
Carbon source: Glucose, glycerol, or molasses Temperature 30–37°C Cell Removal
Centrifugation or filtration to separate cells from the culture supernatant
Minerals: Mn²⁺, Mg²⁺, Ca²⁺ (often essential for PGA synthesis) pH 6.5–7.2 (controlled automatically) PGA Precipitation
Add ethanol or isopropanol (1:1 or 1:2 ratio)
PGA precipitates as a gel or fibrous mass
pH: Maintain at 6.5–7.5 (acidic conditions inhibit yield) Aeration rate ≥1 vvm (volume air/volume medium/minute) Washing
With alcohol or water to remove impurities (e.g., proteins, residual sugars)
Aeration: High oxygen demand → controlled via agitation and sparging Time 48–72 hours (batch) Purification
Optional ultrafiltration or dialysis (to improve cosmetic-grade purity)
Activated carbon treatment if color reduction is required
Yield ~20–80 g/L (can go higher in optimized fed-batch) Drying
Spray-drying or lyophilization (freeze-drying) for powder form
