Pichia pastoris is widely used to produce a variety of exogenous recombinant proteins, and as a model organism to study peroxisome biogenesis as well as methanol assimilation. However, it also has some disadvantages, especially for gene modification, the efficiency of homologous recombination is low. Since targeted gene editing requires several hundred or more homologous overhanging base pairs, the editing efficiency is only 0.1% ~ 30%. Therefore, implementing a highly efficient and convenient gene editing tool is urgently needed.
